Trypticase soy yeast extract medium
Trypticase Soy Broth (BD 211768) 30.0 g, Yeast extract 3.0 g, Agar 15.0 g, Distilled water ad 1000 ml. Adjust for final pH 7.0 - 7.2.
Autoclave at 121 ºC for 15 minutes.
Tryptose 10 g, Meat extract 5 g, Yeast extract 5 g, Proteose peptone 10 g, Sodium chloride 5g, Agar 20 g, H2O ad 1000 ml, Final pH
7.2 at 25 ºC. May be supplemented with L-Cysteine HCl 0.25 g/l.
Following germs should have a good growth at 35 °C in 24-48 hours:
Staphylococcus aureus ATCC 25923, Streptococcus pyogenes ATCC 19615.
Pancreatic digest of casein 15 g, Enzymatic digest of soya bean 10 g, NaCl 5 g, Agar
15 g, Distilled water ad 1000ml; adjust pH to 7.3 ± 0.2.
Sterilize by autoclaving at 121 ºC for 15 minutes.
Medium may also be used as a blood agar base. Melt, cool to 45-50 ºC then add 5%
sheep or bovine blood.
Medium is used for the growth of a large variety of microorganisms. May also be used for colony counting.
