COMPOSITION
Sauton's medium
KH2PO4 0.5 g; MgSO4  * 7H2O, 0.5 g; citric acid, 2.0 g; ferric ammonium citrate, 0.05
g; glycerol, 60 ml; sodium glutamate, 4.0 g; distilled water ad 1 L.
Dissolve the reagents, except glycerol, in 300 ml of water using heat. Add glycerol
then bring the volume to 1 L with distilled water. Adjust pH to 7.4 with 1 M NaOH.
Autoclave at 121 ºC for 15 min.


NOTE
Adding ZnSO
4 * 7H2O, 1% solution 1ml/L of medium improves growth of some
Mycobacteria.


Variants
Sautons Fluid Medium Base by HiMedia
ferric ammonium citrate (brown) 0.0167 g, L-asparagine 1.33 g, citric acid 0.66 g,
MgSO
4 * 7H2O 0.166 g, dipotassium hydrogen phosphate 0.177 g, sodium
dihydrogen phosphate 0.056 g, sodium chloride 0.035 g, polysorbate 80 0.833 g,
distilled water ad 1000 ml.
Adjust pH (at 25 ºC) to 7.2±0.2. Sterilize by autoclaving at 121 ºC for 15 minutes.


PROCEDURE
Inoculate a loopful of Mycobacterium culture and place it on medium surface. Do not
let the culture sunk (admin note).


QUALITY CONTROL
Mycobacterium smegmatis ATCC 14468: good growth;
Mycobacterium tuberculosis H37RV(25618): good growth.
