Potato starch / potato flour 30.0 g, L-asparagine 3.6 g, KH2PO4 2.4 g; MgSO4 *
7H2O, 0.24 g; magnesium citrate 0.6 g, glycerol 12 ml.
Dissolve the salts and asparagine in water. Add glycerol and potato flour and keep
in a boiling water bath with continuous stirring until it reaches a pasty consistency.
Cool to 56 ºC in a water bath and keep the mixture at this temperature for one hour.
Prepare malachite green solution 2 g in 100 ml distilled water. Autoclave at 121 ºC
for 20 min.
Prepare egg suspension from 20-22 fresh chicken eggs to obtain one liter of
suspension. Sterilize the eggs surface, harvest the yolks and egg whites then mix
Final medium: use salts & potato starch solution 600 ml, malachite green solution
20 ml, and egg suspension 1000 ml. Homogenize them then filter the mixture
aseptically through several layers of gauze.
Distribute the medium in tubes (6-8 ml / tube). Heat to 85 ºC for 50 minutes in a
declive position to create a slant. When heated, the egg albumin coagulates,
providing a solid surface for inoculation.

solid egg-based medium used for the isolation and cultivation of Mycobacterium
L-asparagine and potato flour are sources of nitrogen and vitamins. Monopotassium
phosphate and magnesium sulfate act as buffers and support growth. Malachite
green prevents the growth of most contaminants that survive decontamination of the
inoculum, while encouraging the growth of mycobacteria. The egg suspension
provides fatty acid and proteins necessary for the metabolism of mycobacteria and
the solid state of medium when coagulated.
