DESCRIPTION
Some bacteria are able to hydrolyze carboxymethylcellulose. The cellulases produced by bacteria will lower the viscosity of the
medium (a thick syrup of cellulose in a minimal medium has a high viscosity).
MEDIUM PREPARATION
Na-taurocholate ............................................................................... 5 g
NH4H2PO4 ......................................................................................... 1 g
MgSO4 * 7H2O ............................................................................... 0.2 g
KCl .................................................................................................. 0.2 g
Bromthymolblue ........................................................................ 0.05 g
CaCO3 .............................................................................................. 3 g
Carboxymethylcellulose ................................................................ 5 g
PROCEDURE
Harvest a well isolated colony and inoculate a medium tube. Incubate at 37 °C, min.
24 hours. Positive control: Erwinia chrysanthemi, negative control: Erwinia carotovora
subsp. carotovora
RESULTS
Check viscosity by comparioson with a negative control. Reaction is positive if the inoculated medium is more fluid than the control
tube.
Check culture for the yellowish-green color produced by the cellulose degradation. The color change is not obligatory.
