Phylum Firmicutes, Class Bacilli, Order Lactobacillales, Family Lactobacillaceae, Genus Lactobacillus [Group A lactobacilli (obligately
homofermentative), Lactobacillus delbrueckii phylogenetic-group], Lactobacillus johnsonii Fujisawa, Benno, Yaeshima and Mitsuoka
Synonym: Lactobacillus acidophilus group B-2 of Johnson et al. (1980).
Gram-positive, short to long rods (0.5-1.5 x 1.5-10 μm) occurring singly, in pairs and
in short chains. Nonspore-forming.
Colonies on BL (glucose-blood liver) agar (2 days incubation under 100% CO2 at 37
ºC) are 0.5-2.0 mm in diameter, circular to slightly irregular, grayish brown to reddish
brown, and rough. Facultatively anaerobic. No growth at 15 ºC. Can grow at 45 ºC and
in 4% NaCl medium.
Isolated from human blood and feces of chicken, mice, calf, horse and pig.

Obligately homofermentative (hexoses are fermented almost exclusively to lactic acid by the Embden-Meyerhof pathway; pentoses or
gluconate are not fermented).
Positive results for acid and alkaline phosphatase (weak reaction), alpha-glucosidase (weak reaction), beta-glucosidase,
beta-galactosidase, fermentation of: amygdalin, cellobiose, esculin, fructose, galactose, glucose (without gas production), lactose,
maltose, mannose, salicin, and sucrose.
Negative results for arginine dihydrolase, catalase, esterase (C4), esterase lipase (C8), lipase (C14), alpha-fucosidase,
alpha-galactosidase, beta-glucuronidase, N-acetyl-beta-glucosaminidase, indole production, leucine arylamidase, lysine
decarboxylase, alpha-manosidase, nitrate reduction, ornithine decarboxylase, oxidase, trypsin, alpha-chymotrypsin, urease, valine
arylamidase, Voges-Proskauer test, fermentation of: arabinose, mannitol, melezitose, rhamnose, ribose, sorbitol, and xylose.
Variable fermentation of melibiose, raffinose, starch, and trehalose.
